It’s happened to me in both D and manual mode matey. And the reason for your problem is in fact this 'Shift Lock'. The master tech at my dealership, and also the techs at VW have said its a safety feature built into the DSG box to protect itself. This is why i am almost certain there must be some faulty sensor, which would lead back to the mecatronics unit.

Update:
Dealer is still waiting on the replacement Mechatronics box!
Meanwhile the DSG has dropped into 'neutral-in-D' again. This time I tried both 'S' mode and manual selection and the 'D', 'S' or gear number blinked and there was no drive. Restarting he engine in 'N' , which I did while still moving cleared it...

Further Update:
After having to wait 10 days for the new mechatronics box to be delivered to the dealer, this was fitted today.
All seem fine thus far. Be assured I will report any further problems. This work was done just 25 months after delivery and was performed free of charge. They even gave me a free Touran TSI for the day which was even better equipped than my Polo - well it had the parking camera.
So I am at least one happy VW customer tonight...

Glad to hear you got yours sorted mate. Still no luck with mine, wish my dealership would wake up and sort this problem out.

This happened tonight. 2nd time i have ever driven my Polo GTI after collecting it on Monday. Was in manual, pulling up to a set of traffic lights. Car went into first, and then the lights changed. I pressed the accelerator and there was no drive at all. I was stranded in the middle of a set of traffic lights. Had to restart the engine to clear the fault. Very dangerous.
Shall be calling my dealership first thing tomorrow.

I've had the same issue a few times with my DSG as well. It seems to clear itself at the moment though. What i did get though was a knocking sound when the gears were changing down (not sure if it's related!) from the passenger side of the car. I mentioned this as they were looking into my oil issue and they're now replacing the whole gearbox.

I have now driven a few 100 kms since the new Mechatronics box and I am please to say that the gearbox is behaving.
It is however no different than before and will still get confused by roundabouts, in slowing down for one and them accelerating off while still moving. This still induces the DSG jump.
The Garage sent me the bill for replacement. All zeros, accept for replacing ½ litre of oil and ½ litre of radiator fluid - about £8. The Machatronics unit would have been £1660!!
